P450-Glo™ Assay Systems
P450冷光前驅受質(proluciferin)當被P450剪切後,會釋放出aminoluciferin,接著會與UltraGlo™ Luciferase作用產生極亮冷光,且因為UltraGlo™ Luciferase對冷光酶抑制劑的耐受性高,因此可減少偽陽性結果。
冷光強度與P450活性成正比,長效冷光可維持2小時。,P450-Glo™ Screening System相對於Assay System的差異為,可提供如P450酵素標準品、Buffer等完整的試劑組合,實驗不需再製備其他試劑即可完成分析,提供快速、準確,方便的P450活性測試平台。

Pgp-GloTM Assay System
原理簡介
偵測P-Glycoprotein (Pgp) ATPase的長效冷光、均質試劑,可用於偵測Pgp substrates和inhibitors。P-glycoprotein又稱為Pgp、MDR1或 ABCB1,約170 kDa,是細胞膜表面的幫浦,排出細胞內的drug compound等毒性物質以保護細胞。本產品偵測compounds對於cell membrane fraction中的recombinant human Pgp的效應。 先將ATP與Pgp作用消耗ATP,再加入luciferase試劑與剩餘的ATP反應產生高亮度、長效冷光。產光與Pgp活性成反比。 若樣品中含的compound刺激Pgp ATPase,產光量會顯著的低於untreated samples。
UGT-Glo™ Assay
測UDP glucuronosyltransferase (UGT) activity的冷光分析試劑。使用UGT-Glo Multienzyme substrate含兩種受質,
可偵測多種樣品來源或是多種UGT isozyme。長效冷光,操作簡單方便,產光與UGT activity成反比。

MAO-Glo™ Assay
長效冷光型的monoamine oxidase (MAO)分析試劑,可偵測合成或天然來源的MAO A和MAO B活性。產品內含luminogenic MAO substrate,受樣品中的MAO催化為methyl ester luciferin,再與試劑中esterase、Luciferase作用產生極亮長效冷光,產光量與MAO活性成正比,冷光可持續約5小時。
